Homozygous screening

After multiple times of planting and screening, ~12,000 homozygous lines were obtained, allowing for the screening, phenotype recording and analysis with the recessive genes.

T2 generation seeds were used for homozygous line screening. For one primary transformant, we selected 30 seeds of 12 independent T1 generation plants respectively, and cultured in water with 30 mg/L hygromycin. The line(s) which all seeds could germinate was considered as homozygous line.
